چکیده

Methods nCor a 8 was purified from raw hazelnuts by precipitation and chromatographical steps. Subsequently, the protein was identified by N-terminal sequencing and mass spectrometry. Stability of the protein was investigated by simulated gastric and duodenal digestion assays. Hazelnut lipids were extracted employing the hexane method. Finally, IgE binding activity of native and reduced/alkylated nCor a 8 was tested in ELISA using sera from 6 hazelnut allergic patients.

Structural and Functional Characterization of the Hazelnut Allergen Cor a 8

Nonspecific lipid transfer proteins (nsLTPs) are basic proteins, stabilized by four disulfide bonds, and are expressed throughout the plant kingdom. These proteins are also known as important allergens in fruits and tree nuts. In this study, the nsLTP from hazelnuts, Cor a 8, was purified and its crystal structure determined. The protein is stable at low pH and refolds after thermal denaturatio...

Hazelnut (Corylus avellana) vicilin Cor a 11: molecular characterization of a glycoprotein and its allergenic activity.

In Europe, hazelnuts (Corylus avellana) are a frequent cause of food allergies. Several important hazelnut allergens have been previously identified and characterized. Specific N-glycans are known to induce strong IgE responses of uncertain clinical relevance, but so far the allergenic potential of glycoproteins from hazelnut has not been investigated. The aim of the study was the molecular cha...

Is Microarray Analysis Really Useful and Sufficient to Diagnose Nut Allergy in the Mediterranean Area?

BACKGROUND Component-based diagnosis on multiplex platforms is widely used in food allergy but its clinical performance has not been evaluated in nut allergy. OBJECTIVE To assess the diagnostic performance of a commercial protein microarray in the determination of specific IgE (sIgE) in peanut, hazelnut, and walnut allergy. METHODS sIgE was measured in 36 peanut-allergic, 36 hazelnut-allerg...

Predictive values of component-specific IgE for the outcome of peanut and hazelnut food challenges in children.

BACKGROUND Oral challenges are the gold standard in food allergy diagnostic, but time-consuming. Aim of the study was to investigate the role of peanut- and hazelnut-component-specific IgE in the diagnostics of peanut and hazelnut allergy and to identify cutoff levels to make some challenges superfluous. METHODS In a prospective and multicenter study, children with suspected peanut or hazelnu...

Performance of component resolved microarray diagnosis in nuts allergy

Performance of commercially available component resolved microarray has not been widely evaluated in food allergy. Nut allergy is a prevalent and increasing food allergy in which component resolved diagnosis is crucial. In the context of a multicentric study on pollen and vegetable food allergy in Spain, we enrolled patients (n=100) who had suffered at least two episodes of immediate symptoms a...
